Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. The specificity of this monoclonal antibody to its intended target was validated by HuProt™Array, containing more than 19,000, full-length human proteins. DNMT3A is required for genome-wide de novo methylation and is essential for the establishment of DNA methylation patterns during development. DNA methylation is coordinated with methylation of histones. It modifies DNA in a non-processive manner and also methylates non-CpG sites. May preferentially methylate DNA linker between 2 nucleosomal cores and is inhibited by histone H1. Plays a role in paternal and maternal imprinting. Calponin is a calmodulin, F-actin and tropomyosin binding protein, which is thought to be involved in the regulation of smooth muscle contraction. Calponin expression is restricted to smooth muscle cells and has been shown to be a marker of the differentiated (contractile) phenotype of developing smooth muscle. Multiple isoelectric variants of calponin have been identified, however only two molecular weight isoforms exist, a 34 kDa form and a 29 kDa form. Expression of the 29 kDa form, I-calponin, is primarily restricted to muscle of the urogenital tract, whereas the higher molecular weight variant has been demonstrated in vascular and visceral smooth muscle. Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker. It shows antiproliferative activity and has been proposed to play a role in normal cell cycle regulation, replicative senescence, cellular immortalization, and tumor suppression. Thyroglobulin is a 660 kDa dimeric pre-protein with multiple glycosylation sites. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine. Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation. The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary and follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor. This MAb recognizes a protein of 56 kDa, identified as cytokeratin 6 (CK6). In humans, multiple isoforms of Cytokeratin 6 (6A-6F), encoded by several highly homologous genes, have distinct tissue expression patterns, and Cytokeratin 6A is the dominant form in epithelial tissue. The gene encoding human Cytokeratin 6A maps to chromosome 12q13, and mutations in this gene are linked to several inheritable hair and skin pathologies. Keratins 6 and 16 are expressed in keratinocytes, which are undergoing rapid turnover in the suprabasal region (also known as hyper-proliferation-related keratins). Keratin 6 is found in hair follicles, suprabasal cells of a variety of internal stratified epithelia, in epidermis, in both normal and hyper-proliferative situations. This antibody recognizes a protein of 39 kDa, which is identified as Aurora kinase B (or Aurora B). Aurora B is a serine/threonine protein kinase and component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. It is important for accurate chromosome segregation, cytokinesis, protein localization to the centromere and kinetochore, correct microtubule-kinetochore attachment, and regulation of the mitotic checkpoint. Aurora B forms a tight complex with inner centrosome protein and survivin. Inactivation of any of these proteins causes similar defects in chromosome segregation. The intestine-specific transcription factors CDX1 and CDX2 are important for directing intestinal development, differentiation, proliferation and maintenance of the intestinal phenotype. CDX2 protein expression has been seen in GI carcinomas. Anti-CDX2 has been useful to establish GI origin of metastatic adenocarcinomas and carcinoids and is especially useful to distinguish metastatic colorectal adenocarcinoma from lung adenocarcinoma. However, mucinous carcinomas of the ovary also express CDX2 protein. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Spectrin is an actin crosslinking and molecular scaffold protein that links the plasma membrane to the actin cytoskeleton, and functions in the determination of cell shape, arrangement of transmembrane proteins, and organization of organelles. Spectrins function as membrane organizers and stabilizers, composed of non-homologous α andβ chains, which aggregate side-to-side in an anti-parallel fashion to form dimers, tetramers and higher polymers. Spectrin α I and spectrinβ I are present in erythrocytes, whereas spectrin α II (also designated fodrin α) and spectrinβ II (also designated fodrinβ) are present in other somatic cells. This MAb recognizes a protein of 130 kDa, identified as Ksp-cadherin. Cadherins form a superfamily of related glycoproteins that mediate calcium-dependent cell adhesion and transmit signals from the extracellular matrix to the cytoplasm. Cadherins have been implicated in embryogenesis, tissue morphogenesis, tissue structure maintenance, cell polarization, neoplastic invasiveness and metastasis, and membrane transport. It is suggested that Ksp-cadherin is a marker for terminal differentiation of the basolateral membranes of renal tubular epithelial cells. Within the kidney, Ksp-Cadherin is found exclusively in the basolateral membrane of renal tubular epithelial cells and collecting duct cells, and not in glomeruli, renal interstitial cells, or blood vessels.
